Abnormal sleep in the pathogenesis of Alzheimer Disease


Alzheimer disease is a progressive , irreversible disorder of brain that destroys memory and thinking ability of a person and in later stages ability to perform a simple task . Alzheimer's also lead to Dementia which is loss of cognitive functioning - thinking , reasoning , remembering and interfere with person's daily life activities. Abnormal clumps and tangled bundles of fiber are found in the brain of an infected person. The connection between the neurons are also a challenging complication. The damage in brain starts much before the apperence of symptoms.

APP is a surface protein of brain cell and sensitive to many signals. If it is cleaved into four parts it lead to generation of new synapses and formation ofnew brain cell . But if it is broken into four parts it lead to cell death. In normal cases, these two process are balanced . In Alzheimer disease the destruction process predominate the creation process. The reason behind we learn new processes & concept and forget the old unnecessary things are based on these two pathways.


Disturbed sleep increases the risk of occurence of Alzheimer disease. Worse sleep quality , nocturnal wandering , nocturnal awakening and daytime sleepness results in the accumulation of beta-amyloid and tau .

Beta-amyloid is one of the four pieces when APP is broken into for part and lead to generation of more beta-amyloid initiating a positive feedback loop. It also behave as a signal which determine whether a cell have to generate a new cell or it should start the process of apoptosis (leading to the natural death of cell). The feedback loop works in a bi- directional manner if it is somehow directed toward the formation pathway it can help to decrease the damage in brain.



Neuronal damage is triggered in case of abnormal sleep habits and the 'carcadian pacemaker' (internal biological clock ) is also affected. The nerve signal is inhibited and the day to day process is affected.
